MultiCell High Throughput System

The Multicell High Throughput System is an advanced, fully-automated solution for identification and mechanical phenotype quantification of hundreds of myocytes per hour.

FEATURES

  • Measure at a rate of several hundred cells each hour
  • Set cell locations for further measurements
  • Automated transient marking and rejection in rapid batch analysis
  • Automated cell detection and logging
  • Accurate control of the temperature

Using automated cell finding image analysis technology, the MultiCell HTS system provides high-speed, high-content mechanical myocyte data collecting from hundreds of myocytes per hour. The device incorporates a quick x-y-z position programmable scanning microscope that allows for repeated measurements of the contraction-relaxation function by marking and locating the position, size, focus, and orientation of myocytes. The enclosed microscope also maintains a constant temperature while providing complete access for the installation of perfusion systems and electrical stimulation leads.

Higher throughput is necessary to accommodate speedier drug discovery and a growing desire in basic research toward rapid, large-scale data collecting and processing, even though studies using IonOptix's regular Calcium and Contractility systems have shown to be incredibly helpful for many years. High functional variability in isolated myocytes can lead to inaccurate characterisation of crucial variables used to calculate myocyte dynamics. Myocyte function can be characterized more precisely and confidently when data from a considerably larger number of myocytes are collected. This is a significant improvement in phenotypic quantification in the pharmaceutical industry since it speeds up the evaluation of therapeutic efficacy/toxicity and significantly lowers costs by removing hazardous or unneeded components.

In order to retain the highest degree of accuracy while keeping a positive and realistic experience for users in the lab, every technological aspect of the MultiCell HTS has been carefully taken into account. Batch analysis makes it possible to characterize hundreds of cells quickly and automatically while still identifying all of the crucial variables used in IonWizard's core analysis. Tau calculations, a crucial metric used to define sodium-calcium exchange and SERCA activity, benefit from improved data fitting due to double exponential fitting of calcium re-uptake. MultiCell HTS provides substantially faster acquisition and analysis rates while yet providing the same complete acquisition and analysis that our users have come to expect from an IonOptix system.

MultiCell Lite

As an upgrade to current Calcium and Contractility systems or as a standalone system, MultiCell Lite is a less costly choice. MultiCell Lite, which consists of a motorized stage mounted on an inverted microscope, provides the same software capabilities as MultiCell (repeated measurements, automated cell finding and recording), as well as faster data collection than a conventional Calcium and Contractility system at a lower price than the full MultiCell.

COMPONENTS

MultiCell High Throughput Systems can be tailored to your individual requirements. Depending on your application, systems can incorporate balanced flow perfusion, multi-well or single-well dishes, and excitation light sources and optics that are tailored to any fluorescent probe. Square pixels on the new MyoCam-S3 enable digital rotation, and a sCMOS sensor produces fast frame rates.

Microscope

Select between an inverted microscope for the MultiCell Lite or the enclosed CytoCypher microscope for the full MultiCell system.

Light Source

Your unique research requirements guide the selection of the fluorescence excitation light source, which can be set up for practically any dye or dye combination. Monochromatic LEDs, the extremely quick HyperSwitch, or the more reasonably priced MuStep are all possibilities.

Sensors

The conversion of cellular signal into data occurs within MultiCell systems' sensors. A photomultiplier tube and the high-speed MyoCam-S3 digital camera are used to assess whole-cell fluorescence photometry and cell contractility, respectively.

Cell Stimulator

To give cells a quick stimulation, the field stimulator is a crucial part of excitation-contraction investigations. For complex sequencing and arrhythmic stimulation, use the MyoPacer EP instead of the MyoPacer for straightforward stimulation procedures.

Interfaces

The FSI links the hardware and software components of the MultiCell system, enabling synchronous control of calcium and/or contractility measurements along with inputs and outputs for external hardware.

Cell Chambers and Perfusion

The CytoCypher Balanced Flow Perfusion System can be utilized in conjunction with several dish types and stimulation/perfusion lids.

SOFTWARE

IonWizard smoothly combines new modules for MultiCell capture and automatic cell detection and recording with the well-known calcium and contractility trace recording.

You may execute enhanced sequencing functionality with the new IonWizard 7, making it simple to set up MultiCell and standard experiments by constructing epoch loops. Moreover, cell locations are maintained, and data files can be recovered in the case of a software crash.

The MultiCell system comes with seven modules in addition to IonWizard to make a complete, high-throughput system.
